The novel reductions of HCN to CH4, NH3 and tentatively, small amounts of CH3NH2, and of N3- to NH3 and N2 are demostrated with cell-free extracts of Azotobacter vinclandii and Clostridium pasteurianum. Requirements for the reductions are: (i) extracts of cells grown on N2 (those grown on urea or NH3 are infective); (ii) ATP-generator; and (iii) reductant (Na2S2O4 or in some cases H2 or KBH4) and characteristics of the reductions are: (i) inhibition by either substrate of ATP-dependent H2 evolution from Na2S2O4, but no effect on reductant-dependent ATPase; and (ii) complete inhibition by CO. The rate of electron consumption for reduction of N3- is similar to the rate for reduction of N2 or N2O, while the rate for reduction of HCN is about 30% of the others. |
